That Biggus Dickus scene is definitely one.

 

When he sidles up beside the centurion and says "he has a wife you know...." I lose it.

Bill Murray in Kingpin, especially the bit where the female fan runs onto the lane to hug Big Ern and he escorts her into the arms of the waiting security by holding her tit. His hair on the final ball gets me as well. every time.

 

Love the bit in that where they're bowling at the rich guys house, and they start scrapping and the lights go out. One of the heavies thinks he's got Woody Harrelson round the neck, and says "I'll poke your eyes out you bald bastard". When the light go on he's holding a bowling ball under his arm with his fingers in the holes. Superb.

EDIT: I'm trying to avoid outright comedies because they're meant to be funny. You might as well post the entire movie rather than specific scenes when choosing stuff like Life Of Brian and Airplane!

I the cinema, watching Prometheus, I genuinely laughed out loud at them running away from that massive doughnut in a straight line. I think I may have done the same when Rafe Spall was going "oochy coochy coo" to an alien lifeform.

Don't think I can find a video of it, God knows Youtube wouldn't have it. But if anyone knows what I'm talking about, the first scene with "Lolita" in Broken Flowers with Bill Murray is hilarious. Watching that in the theatre with other people's reactions around me was outstanding.

Was watching National Lampoons Xmas Vacation at the weekend & there are a few excellent scenes in that.

The film is full of them. The passengers queuing up to slap the hysterical woman, jive talk, the crash positions, "Joey, have you ever seen a grown man naked?", etc.
